ABSTRACT

Irritable bowel syndrome ( IBS ) is one of most common gastrointestinal dysfunction diseases in worldwide,it often causes recurrent abdominal pain,bowel habit change ( diarrhea or constipation) ,abnormal defecate character and so on. These symptoms occur recurrently and persistently,which seriously affects the patient's daily life. How to make a rational treatment plan for IBS patients is an urgent problem to be solved. This article reviews the recent advances in pathogenesis and treatment of IBS.

ABSTRACT

AIM: The experiment was designed to study the effect of compound Danshen dripping pills (DSDP) on myocardium with anoxin/reoxygenation. METHODS: The myocardial anoxin/reoxygenation model was made in perfused isolated rat heart. DSDP and isosorbide dinitrate (ID) were given at the time of pre-perfusion and reperfusion, then HPLC and H-600 electron microscope were used to detect the change of high energy phosphate and the ultrastructure of myocardial cell. RESULTS: ① The contents of AMP, ADP, ATP and AN in myocardium in only anoxin/reoxygenation group were obviously lower than those in the control group (P0.05). ③ In the groups with ID, the contents of AMP, ADP, ATP and AN were distinctively lower than those in the control group (P

ABSTRACT

AIM: To examine the effect of compound salvae-dropping-pill (CSDP) on intracellular free calcium in cultured rat myocardial cells subjected to hypoxia and reoxygenation.METHODS: The Fluo- 3/AM was applied to probe intracellular calcium concentration and the fluorescent intensity was detected using laser confocal microscopy technique.RESULTS: Fluorescent intensity in hypoxia plus CSDP group was significantly lower (1 217 78?312 07) than that of hypoxia group (1 509?508 48), and the Fluorescent intensity of hypoxia/reoxygenation plus CSDP group was also markedly lower (1 567.91?577 61) than that of hypoxia/reoxygenation group (1 617.60?477.53).CONCLUSION: The cultured rat myocardial cells could be effectively protected by administration of CSDP in case of hypoxia and reoxygenation through decreasing the intracellular calcium concentration. ABSTRACT

AIM:To observe the expression of calcium-sensing receptor(CaSR) of rat cardiomyocytes in anoxia-reoxygenation(A/R) injury and CaSR-induced changes of intracellular calcium;involvement of CaSR in apoptosis and relevant signaling transduction pathway.METHODS:The A/R injury was remodeled in vitro;CaSR,caspase 3 and caspase 9 were deteced by Western blotting.LSCM was used to observe changes of intracellular calcium during reperfusion with or without CaSR agonist.Morphological changes in different groups were observed by light microscopes.Apoptotic cells were measured by TUNEL assay.RESULTS:By LSCM,it was found that the intracellular calcium was significantly increased during reperfusion both in A/R and activator group.Severe injury was found by HE staining in the above two groups,the number of apoptotic cells was significantly increased according to TUNEL assay.The expression of CaSR,caspase 3 and caspase 9 was significantly increased in A/R group and activator group compared with control.CONCLUSION:CaSR is involved in intracellular calcium overload in A/R cardiomyocyte,which can accelerate apoptosis during A/R.

ABSTRACT

AIM: To observe the effect of Dan Shen Pill on expression of Fas/FasL proteins in cultured neonatal rat cardiomyocytes during hypoxia and reoxygenation. METHODS: The neonatal rat cardiomyocytes were isolated and cultured in normal condition for 24 hours before hypoxia and reoxygenation experiments were conducted. The expressions of Fas/FasL proteins in cultured neonatal rat cardiomyocytes were determined by immunohistochemistry and positive expression index (PEI) was calculated. RESULTS: (1) The PEI of Fas/FasL proteins in hypoxia group was higher, but the PEI in Dan Shen Pill treated group was significantly lower than that in control. (2) Under the condition of hypoxia for 30 min then reoxygenation for 4.5 h or 10.5 h, the PEI of Fas/FasL oprotein in cultured neonatal rat cardiomyocytes was also much higher than that in control. However, in the presence of Dan Shen Pill, PEI reduced remarkably compared to cells in the absence of Dan Shen Pill under the same hypoxia and reoxygenation conditions. CONCLUSION: The results indicated that the enhancement of Fas/FasL protein expression emerged in the hypoxia and reoxygenation injury in cultured neonatal rat cardiomyocytes. Dan Shen Pill treatment reduced apoptosis during hypoxia and reoxygenation by decreasing the Fas/FasL protein expression.

ABSTRACT

AIM: To investigate the role of heat-shock protein 70(HSP_(70)) in the protection of myocardial cells against ischemic injury. METHODS: Myocardial cells were cultured in vitro. HSP_(70) was induced by hyperthermia.H_2O_2-injured myocardial cells were divided into different groups. Flow cytometry, DNA Ladder and biochemistry methods were employed to observe the myocardial cells of different groups. RESULTS: Immunohistochemistry showed hyperthermia induced the up-regulation of HSP_(70) in myocardial cells. Apoptotic rate, activity analysis of cytochrome C and succinic dehydrogenase in H_2O_2-injuried and HSP_(70)-protected groups were obviously different. Electron micrograph shomed hyperthermia alliviated myocardial cell injury induced by H_2O_2. CONCLUSION: HSP_(70) delays apoptosis and protects against H_2O_2-induced myocardial cell injury.
